No, the joke is that Cheerilee is telling a penis joke to a group of underage fillies.

Y’know, sad thing is, the current voice of Dr. Robotnik/Eggman (Mike Pollock) after doing a request to say this line, (Sorry the only video I could find of it) after he found out later the context/joke behind it stated he was never going to do fan-request voices again…
 
Meanwhile, after Tara Strong found out what “Melting Pot of Friendship” was from, (Ghost of TCR getting prank-called/trolled) she didn’t mind, and thought it was funny, in fact. She still does voice requests to. (If only Mike was a better sport…)
