Merely proof that their bodies develop over time, perhaps to a state where the body is at its peak. Purely speculation at this time, but Cadence WAS recognized as “The Crystal Princess”. And in those episodes the existence of a Crystal Queen was mentioned.
 
It could be that at the time Sombra took over, Cadence, who was a foal or slightly younger than her teenage years at the time, was sent to Canterlot to live in safety. Remember the Crystal Ponies lost a thousand years and felt as if it were only yesterday, so they’d have a pretty clear memory of what she still looked like.

Yes, there is. Cadance visibly aged between the flashback and the rest of the episode, so she’s not immortal. The jury’s still out on Twilight, but since she doesn’t have the mane of the sisters, it’s safe to say she’s not immortal either.

OP, listen to me: You will see a lot of your family and friends die. In fact you may see all of them die. You don’t need to be immortal to see people you care die.
