Hey,4 years too late but the ‘joke’ has nothing to do with hetero/homo leanings. In the Hearts and Hooves day episode Twist gives a card to Truffle Shuffle, and you see Tootsie Flute looking disappointed because she was going to give a card to Twist.

So it’s simply about making Tootsie happy
