I don’t think so… When she was banished, she was not “on” the moon per se - she was “the” moon. That way enviromental conditions wasn’t much of a problem, and she probobly hadn’t retained consciousness - not body consciousness. The whole banishment was like a really long sleep (or anabiosis).  
This situation is quite different, although I really suppose that even if she dies from exposure, she will be reborn in a new physical shell. Because she has a place in the world, a task to fulfill, and most importantly, an imprint in the minds of people. She and Celestia are akin to Endless from “Sandman” - not so much gods, as physical maniifestations of certain ideas. New Luna won’t retain any memories of the previous one, however much of the physiacal and mental traits will be similar (because that’s how she is remembered).  
That’s me headcanon (one of the many, lol) anyway.

She seals it up with telekenesis and returns to shuttle asap.

Whatever you do, don’t hold your breath. Holding breath in space will kill you instantly and horribly. You can survive longer by releasing all of the air from the lungs.

 

Eh, she’ll be fine. If she can stand a 1000 years in the vacuum of the moon she can stand a few minutes without air.
