Can you imagine having your entire gender put into one character? It’s not like most cartoon shows from the 90s~2010s had girl characters that were just “the girl” of the group. Not at all.
And a show aimed at mainly girls having mainly girl characters?! No way!

Spike wasn’t really one of the mane 6 in G4. He felt like a bit of an outsider to the actual main cast, a sidekick at best but nothing more. This wasn’t helped by Spike being much younger than the mane 6 which made him have a different dynamic than if he had been older.
Hitch has a lot more potential to really explore that which Spike was missing. I mean, I just imagine this same joke in the image being made between the mane 6 and Spike, and it doesn’t have anywhere as much punch as this does because we now know Hitch is meant to be part of the main cast group. Which is why him being “excluded” on the basis of his gender alone despite being on the main cast is funnier.

Did you mean lack in the sense of “main”?
Spike is not fully a part of Mane six, Student six has 2 males, but aren’t mains.
