The author confirmed it’s not her mum because it’s not Trixie. The first draft of the script addressed Trixie by name but editorial had a problem with that. Then the artist just put her in independently.
 
I mean comics aren’t canon anyway but it’s sad that one of the few good things from the comics were rendered even further non canon.

This just means we might get a Trixie parents episode which would unfortunately just be further confirmation that Trixie’s mum from the comics isn’t canon.

The rule of thumb is that the comics are canon until they’re not. Lacking other evidence to the contrary, this appears to be Trixie’s mom.

Basically, writer wanted to confirm it was Trixie’s mom. Hasbro nixed the idea and said to take her out. But the artist didn’t get the memo, so she drew her anyways.

Look at her cutie-mark, she is undeniably related to Trixie. I’d accept sister, but I expect mother. And this Jack Pot, he too could be a brother, or a father. We’ve gotten it wrong in the past assuming parent and child where it was really siblings. But we’ve also gotten it right. It’s gotta be one of the two for sure.

In the comic, she’s just in this one panel here. The context is that Twilight’s parents are dropping filly Twilight off at Princess Celestia’s School for Gifted Unicorns. So the assumption is this is Trixie’s mom. Granted, she could be another relative or just a caretaker, but that’s all the info we have.
 
As for Jack Pot here, the only thing linking him to Trixie is that she looks like a carbon copy of him, and they’re both in the same line of work. He kind of acts like her, too. So, even if they’re not trying to imply it, it sure as heck LOOKS like he could be her dad. I’m certainly not going to argue against that head-canon.
