Maybe, but I think a neurotic, distrustful, and suspicious (occcasionally bordering on paranoid) villain who learns to open up, to trust other people, to accept help instead of trying to do everything on her own, would fit her better. Star Swirl, according to Celestia “did not understand friendship like [Princess Twilight does]”. Combine that with him bringing knowledge of another world, the magical threats in it, and the path(s) between that world and the world Sci-Twi lives in, you can see how the girl might wind up with a siege mentality.

Interesting, but those are two people she*Twilight) HAD to know. She’s lucky she got along with them and became friends with them(because Twilight is a great friend, even if she didn’t know it yet). Her hardcore studies and anti-social behaviour seemed to make her not want to make friends with others. I think of it like a kid who’s been home schooled. They can be perfectly nice to the people they always knew, but aren’t great at making new friendships themselves, or so they think at least.  
Also, she isn’t a very compelling villain for the type of show this is, what’s the worst she could do that she could do in the show with that motive?  
a) Yes, a very popular element we all hope to see :)  
b) I dunno, it’s fanon that’s it’s a dumping ground, considering as we know that only the Sirens were sent there….how about she has concern for her home dimension that it’s only when Equestria is involved that only Equestria can handle it? She would want to close the portal forever to prevent further misfortune for her world(the shows climactic answer to that would be, ‘what good is a world without friendship?’  
c). Again, I dunno. They used it(Magic) to save the world twice, and it only manifests itself as cute ears and longer hair…she could get irked that they didn’t research themselves, but that’s it, I think :p

Wait, I had a new idea after mulling it over a bit. In her flashback with Shining Armor, Twilight seemed pretty normal, and she’s always spoken of Cadance pretty glowingly, so what if Twilight only became cold and friendless a few years into her intense apprenticeship to the princess?
 
Because of this, human Twilight is capable of making friends, and because of it has an element themed team to fight regular Twilight with?
 
It’d still be super easy to make her a villain, since she could a) want to discover Equestria in the name of science b) be ticked at Equestria for using her planet an a dumping ground and/or c) be ticked at the human!elements for wasting their magic gifts.

Hmmm…Scrounge-sempai mentioned Star-Swirl the bearded…perhaps she could have been under his tutelage, and we’ve seen he’s VERY ambitious to learn everything about Magic(science in human world?)  
Taper that in with her teacher(Starswirl) suddenly going missing, or making subtle references that he died, Twilight could want to fulfill the ambition of her only friend.  
You could argue away the time problem by saying time works differently there(Sunset somehow aged from a mid to late teen, yet when Twilight arrived, they appeared the same age despite Twilight being younger than her), but I doubt that is necessary, since I’m sure Celestia and Luna aren’t immortal there, either.

Yeah, you got me there. Still, that’s why I think it would have been cooler. Sunset was kind of a crappy character in the first movie (though I’ll admit they totally turned her around in the sequel and made her pretty cool).
 
True, too, that being 100% devoted to science could make her a villain in her own right. Imagine if Human Twilight was this:
