The cake has no fewer than 21 and no more (again, assuming radial symmetry) than 36 candles, so… possibly maybe.  
Although I do think they meant to imply she’s 21 and just neglected to take the far side of the giant cake into account.  
Or possibly just slapped on a nice-looking number of candles and neglected to take into account bronies’ the Internet’s collective autistic OCD obsession over details. :P

And I thought the “canon arse” in the description was a joke meaning this was the first full plot-shot we’ve got of Luna in the comic so far, since pretty much all other characters have had their butts on display at least once. I missed the first version of this page.

Yeah, I get the metaphor, but their literal ages, after doing the math, the Mane 6 are in their mid to late 20s.
Basically since the CMC are in their tween/early teens and Applejack herself is confirmed to be slightly more than twice Applebloom’s age, that’s a good approximation.
 
Someone counted the candles visible on Dash’s cake in her birthday episode, and said there were 25. So I’d say that’s a good indication of her age, and the others are similar.

That’s correct, the other version had her splotches covering a greater range of her butt, I’m not sure of the exact details that led to the merge though, I assume it’s because this version was meant as a fix, rather than an alternative version.
