Spike wasn’t in Gen 3.5. He was “Master Kenbroath Gilspotten Heathspike” in Gen 3, and instead of being a kid like in Gen 1 or Gen 4, he was a slightly kooky little old man.

I love the idea of Spike being a goofy dragon uncle in general. It’s just too perfect to pass up. To Flurry, to the Cake Twins, to the Mane 6’s future kids.

I guess one could theorize that in Equestrian culture, older siblings have almost parental duties. And the parent unless he/she is very responsible, the parent only really hangs out with the child 50% of the time.
 
That does seem a little cold though. At least dragons are honest about not seeing their children after they get wings. Hopefully the Mane 6 will be better parents.

MLP is notorious for muddling the distinction between “mother and daughter” and “big sister and little sister”. It goes back to Gen 1 with the Baby Ponies, who were filly-sized versions of the main cast. You could interpret them as being either the daughters of the main cast ponies, or their kid sisters. I think Hasbro left it deliberately vague so that little girls could play with either interpretation.
 
Fast-forward to Gen 4, and we see echos of that. Applejack is Apple Bloom’s big sister, but she’s also her de facto mother since their parents are out of the picture. Likewise, Sweetie Belle spends far more time with big sister Rarity than she’s ever shown spending time with their actual parents. So Twilight Sparkle being Spike’s big sister or Spike’s adoptive mother are both valid character interpretations.
