If Scoots’s auntses ever do show up onscreen looking anything like this, it’ll either be because somebody at DHX tipped off PK with some loose design cues, or because they asked her ahead of time to design the characters for them; they don’t just wander out into the fandom and appropriate character designs, it would cause a host of legal problems.

I think there’s a pretty good chance, though, that Hasbro might just create their own original designs for Scootaloo’s aunts. As another user already mentioned below, should Hasbro ever market these two characters in any capacity (which given their track record is rather likely), they would need full control (and thus full copyright) over the designs. Using models of their own creation would immediately grant them that power, whereas intentionally homaging someone else’s work could potentially place them in a hazy legal situation.

Holiday looks like a senior grade math teacher or a secretary.

Given that Pixel has close ties with some of the staff, it wouldn’t surprise me if she got some insight from them as to what the characters looked like.

Ant Lofty here looks like the type who’d make brownies for her nephews and niece every time they visit.

She doesn’t, but she is close enough to the staff that some of them most likely saw this image.
 
If Holiday and Lofty would ever appear in the show, someone in Hasbro or DHX might have to make make the decision to either intentionally homage pixel’s design or intentionally go with something else. Considering the fact that Hasbro needs full copyright control over these characters in case they will make it into the toy line, I suspect it will be the latter.
