So in that instance I guess Twilight would be talking about either science or friendship and she won’t let anyone else talk or really care if anyone’s actually listening to her
 
 
Also, Pissy Twilight looks like she’s actually taking a piss. In a way that a man would.  
I’m pretty sure the artist meant for her to be aggressive looking but it just looks like someone’s being naughty and actually peeing on you. :\

Don't Call Me Brony
The original Japanese for this one is 説教魔, which can be translated literally as “sermon demon”. It just means a person who acts like a know it all and then won’t shut up about it.
