Yes and no. Grant Wood’s original intent was that it was a farmer and wife. The ‘daughter’ interpretation was on the insistence of Wood’s sister, Nan, who was the model for the woman and thought the guy looked too old for her. So officially, it’s a man and his daughter; but the artist when he was drawing it meant for it to be a man and his wife. They’re both entirely defensible interpretations (and if you subscribe to the ‘death of the artist’ philosophy, it is whatever you think it is, no wrong answers).
